摘要
目的:用正交试验法优选返魂草中总黄酮的提取工艺。方法:以50g原药材的总黄酮提取量为评价指标,选择提取用水量(倍)、提取时间(h)、提取次数(次)为考察因素,采用正交试验法确定返魂草中总黄酮的最佳提取工艺,采用分光光度法对干浸膏中总黄酮的含量进行测定,检测波长500nm。结果:最佳提取工艺条件为提取所用水量为12倍,提取次数为3次,回流提取时间为3h,芦丁在9.52～57.12mg.L-1范围内呈良好的线性关系。平均回收率为98.48%,RSD为0.86%。结论:总黄酮正交提取工艺的研究结果比较可靠,含量测定方法简便、准确,可用于返魂草干浸膏中总黄酮的含量测定。
OBJECTIVE To study the optimum extraction process of total flavonoids in Sehecio cannabifolius less.METHODS Using the extraction yield of total flavonoids as the assessment index, some important factors in the experimental orthog onal design, such as volume of extraction water,extracting time, times, were studied in detail. The total flavonoids content ex tracted from Sehecio cannabifalius less was determined by spectrophotometric method at detection wavelength of 500 nm. RESULTS The optimum extracting conditions were as follows： adding 12 fold of extraction reagent, three times, heating for 3 hours. Standard sample of Rutin showed a good linear relationship over the range of 9. 52- 57. 12 mg· L^-1 and the average re covery was 98. 48% and RSD was 0. 86%. CONCLUSION The optimum extradion process of total flavonoids in Sehecio cannabiJolius less was reliable and the determination method is easy and accurate which can be used for the extraction of total fla vonoids in this herb.
