摘要
目的考察用合煎(传统汤剂)、分煎(配方颗粒汤剂)方法制备的鸭掌散汤剂中甘草酸含量的变化,并比较其体外抗菌作用。方法①建立高效液相色谱方法,采用Diamonisl C18柱(250 mm×4.6 mm,5μm),甲醇-0.2 mol/L醋酸胺溶液-冰醋酸(67∶33∶1)为流动相,流速1 ml/min,检测波长250 nm。分析鸭掌散合煎液和分煎液样品中甘草酸的含量变化。②采用稀释法体外测试鸭掌散不同煎液对常见菌种的最低抑菌浓度(MIC)和最低杀菌浓度(MBC)。结果①鸭掌散分煎液中甘草酸的平均含量大于合煎液中甘草酸的平均含量。②鸭掌散分煎液对福氏痢疾杆菌有抑菌作用,而合煎液没有;合煎液对白色葡萄球菌、黄曲霉菌的抑菌作用大于分煎液。结论与传统汤剂比较,鸭掌散配方颗粒汤剂中甘草酸的含量较高,抑菌谱较广。
Objective To investigate the contents of glycyrrhizic acid in combined decoction ( mix the traditional Chinese herbs together, then boil them with water) and the separated decoction (boil the single traditional Chinese herb with water separately, then mix the extracts) of Ya Zhang powder ( composed of Ephedra sinica, Prunus armeniaca and Glycyrrhiza uralensis) and to compare with their anti - bacterial effects in vitro. Methods (1)An HPLC method was established with a Diamonsil C18 column (250 mm×4.6 mm,5 μm), a mobile phase of methanol -0.2mol/Lammonium acetate solution- Acetic acid (67: 33: 1 ), a flow rate of 1 ml · min^-1 and a detection wavelength of 250nm in order to determine the content of glycyrrhizic acid. (2)Minimal bacterial inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) to the common bacteria in different decoctions were measured in vitro by employing dilution method. Results (1)The average content of glycyrrhizic acid in the separated decoction was higher than that of the combined decoction.(2)The separated decoction could display the antibacterial effects to virulence of shigella flexneri, but the combined decoction could not ; to Staphylococcus albus and aspergillus flavus, the antibacterial effect of combined decoction was slightly stronger than that of the separated decoction. Conclusion The content of glycyrrhizic acid in the combined decoction is higher than the separated decoction and the anti -bacterial effect is stronger.
