以FITC-LPS为示踪物对小鼠肠道内毒素移位的观察 被引量:1

摘要 以Pb-Acet和5-HT静注同时LPS灌胃方法形成肠源性内毒素血症动物模型。利用荧光素异硫氰酸盐—内毒素(FITC-LPS)的荧光特性,将其按1/10的比例混入于口灌的LPS中。结果证实,造模3小时后,肝、肺、肾、脑均有较强的荧光发射,说明已有肠道FITC-LPS移位现象发生。本方法具有简单性、可行性、先进性等特点,不失为一种较佳的定性观察LPS移位的实验方法。 Entertoxemia model of mice was made by intravenous injection of Pb Acet and 5 HT with LPS by oral simutaneously.According to the fluorescent character of FITC LPS the model was made by LPS mixed with 10 percent of FITC LPS by oral.Result showed that there were stronger fluorescent emission in liver,lung,kidney and brain 3 hours after the model was made,proving the translocation of FITC LPS from gut.Fluorescent parameters used were Ex495.3nm and Em516.7nm.There was also low fluorescent intensity in normal mice tissues,but it was heterogenous substance from FITC LPS.Optimal parameters were Ex453.8nm and Em517.9nm.
