摘要
目的:观察缺氧性肺动脉高压大鼠循环内皮细胞(CEC)形态学变化。方法:50只大鼠随机分成5组,A组:正常组;B组:缺氧2 w组;C组:缺氧2 w叠加FeCl3组;D组:缺氧4 w组;E组:缺氧4 w叠加FeCl3组。缺氧叠加静注FeCl3模型在每周第3、6 d的缺氧后尾静脉注射0.6%FeCl3。各组分别取血运用多功能显微镜观察CEC的形态学特点。结果:缺氧性肺动脉高压时右室压、肺动脉平均压较正常组升高(P<0.01)。正常组可见少量散在呈不规则多边形的无核、皱缩、折叠或扭曲状CEC;缺氧组CEC数量增加(P<0.01),多有核,胞浆丰富,可见3个以上内皮细胞成片状脱落;缺氧叠加FeCl3组内皮损伤较单纯缺氧组加重。结论:缺氧可致CEC数量增多,缺氧越明显,成片脱落CEC越多,炎症可加重内皮损伤。
Objective:To observe the morphological changes of circulating endothelial cells (CEC) in rats with hypoxic pulmonary hypertension. Methods: 50 male SD rats were randomly divided into 5 groups: group A (normal group), group B (hypoxia for 2 w), group C (hypoxia for 2 w with FeCl3 intravenous injection), group D (hypoxia for 4 w) and group E (hypoxia for 4 w with FeCl3 intravenous injection). Rats in group C and E were injected 0. 6% FeCl3 through caudal vein at the 3rd, 6th day every week. Then, blood samples of all groups were obtained to observe the morphological changes of circulating endothelial cells under Multi functional microscopy. Results:There were more CEC in hypoxia groups than those in normal group (P〈0. 01). These CEC had rich plasma and nucleus. Some slices consisted of over 3 such ceils were found to peel off into the blood. The CEC numbers were markedly increased in hypoxia groups with FeCl3 intravenous injection. Conclusion: Hypoxia can increase CEC numbers. The more markedly the hypoxia is, the more ceils peel into the blood. Inflammation can aggravate the endothelial cell injury.
