脊髓损伤早期细胞凋亡的表达及意义 被引量:3

Expression and significance of early apoptosis in spinal cord injury

摘要 目的:观察大鼠急性脊髓损伤后不同时间细胞凋亡调控基因Bcl-2和Bax的表达。方法:Wistar大鼠56只随机分成7组,将6组制成急性脊髓损伤模型,另一组于伤后0h、4h、8h、24h、48h、72h对其运动功能进行评分,随后采集脊髓标本,分别进行HE染色、TENEL标记和免疫组化检测其凋亡调控基因Bcl-2和Bax的表达。结果:大鼠脊髓损伤后表现运动能力下降。TUNEL染色显示脊髓伤后4h出现凋亡阳性颗粒,伤后8h到达高峰。Bcl-2和Bax都在伤后4h可见阳性表达,Bcl-2表达在24h到达高峰,Bax的表达高峰出现在伤后8h。结论:大鼠脊髓损伤后,除存在细胞坏死外还存在神经细胞凋亡。细胞内Bcl-2与Bax的比例可能调节着凋亡的发生。 Objective:To observe the changes in the expression of apoptosis regulating gene Bcl-2 and Bax at different time points after acute spinal cord injury. Methods. We randomly divided 56 Wistar rats into 7 groups, we made six groups of the 7 groups as acute spinal cord injury model,the other group was used as control. We evaluated motor capacity at Oh, 4h, 8h, 24h, 48h and 72h after injury. The spinal cord was taken and after the HE staining and TUNEL staining,the immunohisto--chemistry methods were used to study the expression of Bcl--2 and Bax. Resuits: After acute spinal cord injury, the motor capacity decreased. TUNEL--positive cells were found at 4h after injury, with a maximum presence at 8h after injury. At 24h after injury,the positive expression of Bcl-2 was maximal,and the positive expression of Bax was maximal at 8h after injury. Conclusion:In addition to necrosis,apoptosis phenomenon presents in rats spinal cord injury. The proportion of Bcl-2 and Bax in ceils may influence upon apoptosis and pathological processes after spinal cord injury.
