摘要
目的:建立以高效液相色谱法测定小儿四维葡钙颗粒中维生素B1、维生素B2含量的方法。方法:色谱柱为ZORBAX Eclipse XDB-C18柱(5μm,250mm×4.6mm)。流动相为0.005mol/L庚烷磺酸钠溶液(含0.5%冰醋酸和0.05%三乙胺)-甲醇(72∶28),柱温30℃,流速为1ml/min,检测波长为260nm,进样量20μl。结果:维生素B1、维生素B2检测浓度的线性范围分别为7.13-22.96μg.mL-1(r=0.9991)、8.12-32.31μg.mL-1(r=0.9993);平均回收率分别为101.1%(RSD=0.7%)、101.4%(RSD=0.6%)。结论:本方法简便准确、灵敏度高、结果可靠,可用于小儿四维葡钙颗粒中维生素B1、维生素B2的含量测定。
Objective Establish the method to determine vitamin B1 and vitamin B2 in Four Vitamin Glucose -Calcium particles by HPLC. Method PE Series 200 type HPLC is used ,which column is ZORBAX Eclipse XDB- C18(5μm, 250mm ×4. 6mm), mobile phase is 0. 005mol/L heptane solution (0.5% acetic acid and 0.05% triethylamine) - methanol (72:28). The detection wavelength was 260nm, flow rate is 1.0mL/min. Assay is external reference method. Result: A good linear correlation was obtained when vitamin Bl's concentration ranged from 7.13μg/mL to 22.96μg/mL( r = 0. 9991 ) and vitamin B2's concentration ranged from 812μg/mL to 32. 31μg/mL(r = 0. 9993 ). Recovery of B1 is 101.1%, RSD is 0. 7% ; Recovery of B2 is 101.4%, RSD is 0. 6%. Conclusion: This method is high - sensitive, separation effect and reproducibility are good, results are reliable, which can be applied to determine vitamin B1 and B2 in Four Vitamin Glucose - Calcium particles for children.
