L-抗坏血酸月桂酸酯的酶法合成、分离及其性质 被引量:12

Enzymatic synthesis,separation and properties of L-ascorbyl laurate

摘要 利用固定化脂肪酶作为催化剂,在丙酮体系中合成L-抗坏血酸月桂酸酯,采用有机溶剂萃取法、硅胶柱分离法和高效液相(HPLC)制备法来纯化,纯度分别达到86.5%、90.4%和95.5%。用质谱(MS)、核磁共振(NMR)对纯品的分子结构进行鉴定,并用Du Nouy拉环法测定出临界胶束浓度(CMC)为0.1mmol/L。采用淬灭DPPH.和ABTS+.自由基方法来衡量抗氧化活性,结果显示L-抗坏血酸和L-抗坏血酸月桂酸酯、棕榈酸酯淬灭醇溶性DPPH.自由基能力具有显著差异,而淬灭水溶性ABTS+.自由基无显著差异,说明L-抗坏血酸月桂酸酯将是一种很有潜力的抗氧化剂。 L-ascorbyl laurate was synthesized by immobilized lipase-catalyze in the acetone system. The separation and purification of the synthesized ester was carried out by organic solvent extraction, silica gel column chromatography, and high performance liquid chromatography (HPLC), respectively. The purities of final products separated by these three methods were 86.5%, 90.4% and 95.5%, respectively. The purified product was subjected to mass spectrometry (MS) and nuclear magnetic resonance (NMR) analysis, while the critical micelle concentration (CMC) determined by Du Nouy ring method was 0.1mmol/L. Finally, the determination of DPPH· and ABTS^+· free radical scavenging ability showed that there was significant differences between DPPH· free radical scavenging ability of L-ascorbyl laurate, L-ascorbyl palmitate and L-ascorbic acid, while no significant difference was found ABTS^+· free radical scavenging ability of these compounds. The results indicated that the L-ascorbyl laurate had great application potential.
