摘要
报道了丝裂霉素明胶微球(MMC-GMS)的研制方法,采用正交设计,优化了制备条件和工艺,采用动态透析法研究了微球的体外释药规律。结果表明:该微球粒径为50~200μm范围内的占微球总数的90.2%;平均粒径为(60.76±9.6)μm,丝裂霉素的包封率为68.4%,载药量为20.8μg/mgGM.释药特性符合Higuchi方程。实验动物灌注微球后病理检查结果表明,灌注后微球成簇堵塞于肝动脉分枝的小动脉,其中大部分已完全堵塞了血管腔。
This paper introduced a method for preparing mitomycin-gelatin microspheres(MMC-GMS).Results:The diameters of 90.2% of the microspheres were between 50 to 200 μm,and the average diameter of the micropheres was(60 76±9 6)μm The drug content in every 1 mg microsphere was 20 8 μg The characteristic of drug release was accordance with Higuchi equation After hepatic artery infusion of the micropheres in experimental animals,the cavities of most of the small branches of the hepatic artery were totally obstructed.
