摘要
目的:观察地骨皮提取液对T2DM大鼠肾组织NF-κB活性,肾脏病理,血清炎症因子IL-6、TNF-α水平的影响,探讨其对T2DM肾脏病变的防治作用及相关机制。方法:应用小剂量STZ加高脂饮食制备T2DM大鼠模型。分为正常对照组(A)、DM无干预组即模型组(B)、罗格列酮干预组(C)、地骨皮醇提高剂量干预组(D)、地骨皮醇提低剂量干预组(E)。用EMSA(电泳迁移率变动分析)方法检测NF-κB活性。结果:药物干预均可不同程度改善肾脏病理,E组大鼠的病理改善略弱于C组,D组肾脏病理改善较显著。与A组相比,B组肾组织NF-κB活性、血清IL-6、TNF-α水平均有显著升高,C、D、E均有升高但明显低于B组,模型组IL-6、TNF-α、尿白蛋白排泄率与NF-κB活性呈正相关(P<0.05)。结论:①通过小剂量STZ加高脂饮食的方法可成功复制T2DM大鼠模型。该模型在第6周已出现糖尿病早期肾脏病变。②T2DM大鼠肾组织中NF-κB活性显著增强。地骨皮干预可减少NF-κB表达,降低血清炎症因子,从而改善肾脏病理、肾功能。
Objective: To observe the effect of extract of Cortex Lycii to T2DM rats' nephridial tissue in term of activity of NF -κB, pathomorphology of the kidney, IL - 6 and TNF -α in serum, so that to investigate the effect and mechanisms of Cortex Lycii to renal lesions in T2DM. Methods: Small dose of streptozotocin(STZ) and high fat diet were used to establish the model of rat with T2DM. Rats were divided into five groups:normal control group (A), diabetic model group (B) ,rosiglitazone group (C), ethanol extracts of Cortex Lycii radicis high dosage group (D)and it's low dosage group (E). EMSA(electrophoretic mobility shift assay)were apphed to evaluate the activity of NF- κB. Results: Improvement of pathomorphology innephridial tissues were observed grout C, D and E, among which grout D had got significant better appearance, and group E was a bit less than group C. Compared to group A, data from group B was significantly higher, including activity of NF - κB in nephridial tissue, IL - 6 and TNF - α in serum. Those data in group C, D and E were higher than group A,but obviously lower than group B. In group B, activity of NF - κB in nephridial tissue positively correlate with IL -6 ,TNF - α, excretory rate of urinary albumin( P 〈 0.05 ). Conclusion: ( 1 ) Model of T2DM rat can be copied by low dose of STZ accompanied with high fat diet. Early renal lesions of diabetes can be observed. (2) Activity of NF -κB increases obviously in nephridial tissue of T2DM rats. Cortex Lycii can reduce the expression of NF -κB,and in further reduce the serum inflammatory factor,and finally improve pathomorphology and function of renal.
