摘要
目的建立高效液相色谱-蒸发光散射检测(HPLC-ELSD)法测定补中益气丸(浓缩丸)的黄芪甲苷含量。方法色谱柱为Diamonsil(钻石)C18柱(200mm×4.6mm,5μm),柱温为30℃,流动相为乙腈-水(38:62),流速为0.8mL/min,ELSD条件为漂移管温度110℃,气体流速3.0L/min。结果黄芪甲苷进样量在2.494～39.904μg范围内与峰面积线性关系良好,r=0.9999,平均加样回收率为97.26%,RSD=1.94%(n=6)。结论所用方法简便快捷,测定结果可靠,可用于补中益气丸的质量控制。
Objective To establish a HPLC- ELSD method to determination of astragaloside IV in Buzhongyiqi Wan. Methods Diamonsil-C18 column （200 mm ×4.6 mm, 5 μm） was employed, and its temperature was 30 ℃. Axetonitrile-water （38：62） was as the mobile phase at the flow rate of 0. 8 mL/min. ELSD inlet heater was set at 110 ℃, and N2 flow rate of 3.0 L/min. Results The calibration curve of astragaloside IV was linear in the range of 2. 494 - 39. 904 μg （ r = 0. 999 9）. The average recovery was 97.26% , RSD = 1.94% （ n = 6）. Conclusion The Method is simple,accurate,reproducible and can be employed for the puality control.
