丙泊酚对大鼠肠缺血再灌注时肝损伤及Bcl-2、Bax蛋白表达的影响 被引量:7

摘要 目的探讨丙泊酚对大鼠肠缺血再灌注时肝损伤及Bcl-2、Bax蛋白表达的影响。方法成年健康Wistar大鼠54只,随机分为3组:假手术组(Sham组)、肠缺血再灌注组(intestine ischemia-reperfusion组,IIR组)、丙泊酚组(propofol组,P组)。各组根据再灌注时间分1,3,6 h三个时相。通过夹闭肠系膜上动脉制作肠缺血再灌注损伤模型,实验结束后即刻取肝左叶做标本。用免疫组化法与医学图像分析检测肝组织Bcl-2与Bax蛋白表达量,用TUNEL技术检测肝细胞凋亡指数(AI)。结果与S组比较,IIR组和P组各时相Bcl-2与Bax蛋白含量、肝细胞AI均显著升高(P<0.05),IIR组各时相Bcl-2/Bax比值均降低(P<0.05),P组各时相Bcl-2/Bax比值均升高(P<0.05)。与IIR组比较,P组各时相Bcl-2蛋白含量、Bcl-2/Bax比值均升高(P<0.05),Bax蛋白含量、肝细胞AI均降低(P<0.05)。IIR组和P组不同时相间肝细胞AI的差异均有统计学意义(P<0.05),而且两组肝细胞AI随再灌注时间的延长而增加(P<0.05),以6 h时相的最高。各组中1,3,6 h三个时相之间Bcl-2、Bax蛋白含量及Bcl-2/Bax比值差异无统计学意义(P>0.05)。结论大鼠肠缺血再灌注后可引发肝损伤,且肝组织Bcl-2与Bax蛋白含量明显升高;丙泊酚可能通过调节Bcl-2、Bax蛋白表达使肝细胞凋亡减轻,从而对大鼠肠缺血再灌注时所引发肝损伤有一定的保护作用。 Objective To investigate the effect of propofol on the expression of Bcl-2 and Bax protein after liver injury induced by intestinal ischemia-reperfusion in rats. Methods Fifty-four Wistar rats were randomly divided into 3 groups( n = 18) : sham group (n = 18), intestine ischemia-reperfusion group(IIR group)and propofol group(P group). The rats were executed in each group after reperfusion for I h, 3 h and 6 h, respectively. The model of liver injury induced by intestinal ischemia-reperfusion was established by blocking the superior mesenteric artery with clap. The left hepatic lobes were sampled at the end of reperfusion. Expression of Bcl-2 and Bax proteins in liver tissues was detected with irnmunohistochemistry and MIAS-2000. Apoptosis index(AI)of cells in liver tissues was detected with terminal deoxynucleotidy-transferase mediated dUTP nick end labeling(TUNEL). Results Expression of Bcl- 2,Bax protein and AI of hepatic cell in IIR group and P group were significantly higher than in Sham group at each time point(P 〈 0.05).Bcl-2/Bax ratio in IIR group was lower than in Sham group at each time point(P〈 0.05),while it was higher in P group than in Sham group(P 〈 0.05). Bcl-2 protein expression and Bcl-2/Bax ratio in P group were higher than in IIR group at each time point(P〈0.05). Bax protein expression and AI of hepatic cell in P group were lower than in IIR group at each time point(P〈 0.05). In IIR group and P group,AI of hepatic cell had significant difference between 3 time points(P〈 0.05). As reperfusion time prolonged, AI of hepatic cell in IIR group and P group increased(P 〈 0.05), highest at hour 6. Expression of Bcl-2, Bax protein and Bcl-2/Bax ratio had no significant difference between 3 time points in 3 groups(P〉0.05). Conclusion Expression of Bd-2 and Bax protein increase after liver injury induced by intestinal ischemia-reperfusion in rats. Propofol can relieve the liver cellular apoptosis by regulating expression of Bcl-2 and Bax protein, which may be one of mechanisms of propofol to protect liver injury induced by intestinal ischemia-reperfusion in rats.
