芎麻滴丸对血管性痴呆大鼠胆碱能神经递质代谢的影响 被引量:5

Effect of Xiongma Drop Pill on the Metabolism of Cholinergic Neurotransmitter in Vascular Dementia Rats

摘要 目的研究芎麻滴丸对血管性痴呆(VD)大鼠胆碱能神经递质代谢的影响,探讨其防治VD的可能机制。方法采用永久性结扎双侧颈总动脉(2-VO)法复制VD大鼠模型,紫外分光光度法检测大鼠脑组织乙酰胆碱(Ach)含量和乙酰胆碱酯酶(AchE)、胆碱乙酰转移酶(ChAT)的活性。结果与假手术组比较,模型组脑组织Ach含量显著降低(P<0.001),AchE活性无显著性差异,ChAT活性显著降低(P<0.05);与模型组比较,芎麻滴丸中、低剂量组及喜得镇组脑组织Ach含量和ChAT活性均显著升高(P<0.05),AchE活性无显著性差异。结论芎麻滴丸防治VD的作用机制可能与其升高VD大鼠脑组织Ach含量和ChAT活性有关。 Objective To investigate the effects of Xiongma Drop Pill (XMDP) on the metabolism of cholinergic neurotrans- mitter in vasculer dementia rats and to explore its possible therapeutic mechanism. Methods The vascular dementia(VD) model of rats was established by permanent occlusion of bilateral common carotid arteries(2- VO). The level of aeetylcholine (Ach), activities of acetyleholinesterase (AchE) and choline aeetyhransferase (CHAT) in brain tissue were detected by UV- spectrophotometer. Results Compared with sham- operation group, the level of Aeh decreased in the model group (P 〈 0. 001), and activity of ChAT decreased ( P 〈 0.05), but the difference of AchE activity was insignificant. Compared with the model group, the middle - dose XMDP and the low - close XMDP increased the level of Ach content and ChAT activity ( P 〈 0. 05), but the difference of AchE activity was insignificant. Conclusion The mechanism of Xiongma Drop Pill in treating vascular dementia may be related to the reduction of Ach level and ChAT activity in brain tissues of vascular dementia rats.
