摘要
目的建立了一种简单的卡马西平萃取方法和高效的反相高液相色谱法。方法血清中卡马西平用乙醚-石油醚(9∶1)单步提取;色谱柱:HYPERSIL ODS(5μm,4.6mm×25mm),流动相:乙睛-0.06mol·L-1NaAc-HAc(pH4.3)=36∶64(内含0.05%的三乙胺)。检测波长285nm,室温,内标:艾司唑仑。结果卡马西平在2-16μg·mL-1范围内呈良好的线性关系(r=0.9998),平均回收率99.91%(n=9,RSD=1.87%)。结论此方法提取步骤少,方法简单快速、专一性强、费用低,适合临床卡马西平的血药浓度监测。
OBJECTIVE A new simple extraction of carbamazepine（CBZ） in serum and an improved method of HPLC were developed. METHODS CBZ was extracted from serum by ethyl ether：petroleum ether（9： 1） and analyzed in a column of HYPERSIL ODS （5μm, 4.6mm×25mm） with a mobile buffer of acetonitrile-0.06 mol·L^-1 NaAc-HAc （pH4.3） （36：64） with 0.05 % Triethylamine. CBZ was detected in 285nm with estazolam as internal standard in normal temperature. RESULTS The linear of CBZ was excellent within the range of 2--16μg · mL^-1 （r = 0. 9998）. The average recoveries were 99.91% （n = 9, RSD = 1.87 % ）. CONCLUSION The method is simple, rapid, sensitive and applied to determine the therapeutic drug monitoring of CBZ.
