MDR_1基因表达与原发上皮性卵巢癌原代细胞体外抗药性关系的研究 被引量:1

摘要  目的 探讨 M D R1 基因表达与原代细胞体外抗药性的关系,为原发上皮性卵巢癌合理化疗提供理论依据。方法 应用半定量逆转录———聚合酶链反应技术和 M T T 法体外药敏试验,检测了37 例原发上皮性卵巢癌原代细胞对临床常用抗癌药物的抗药性及其 M D R1基因表达情况。结果 原发上皮性卵巢癌 M D R1 基因阳性表达率为30 % 。 M D R1 基因表达与 M D R 相关药物耐药有关,与非 M D R 相关药物耐药无关。结论  M D R1 基因表达是原发上皮性卵巢癌抗 M D R 相关药物的主要原因。 Objective To study the relationship between in vitro chemosensitivity of primary epithelial ovarian carcinoma (PEOC) to MDR related drugs and expression of MDR 1 gene.Methods A sensitive semiquantitative reverse transcription-polymerase chain reaction (RT-PCR) assay was used for detection of MDR 1 gene expression in 37 PEOC .Sensitivity of the tumor tissues to the clinical common anticancer agents was examined by means of MTT assay in vitro.Results MDR 1 gene expression was detected in 30% of PEOC analysed.The level of MDR 1 mRNA was not correlated with nonMDR related drugs, but correlated with MDR related drugs.Conclusion These results support the idea that the increased expression of MDR 1 gene is a major cause of resistance to MDR related drugs.
