应激对裸鼠胰腺癌生长的影响 被引量:1

THE EFFECTS OF STRESS ON PANCREATIC CANCER GROWTH IN NUDE MICE

摘要 目的构建裸鼠胰腺癌移植瘤束缚应激模型,探讨应激促进裸鼠胰腺癌生长的机制。方法将20只裸鼠随机分为单纯荷瘤组和荷瘤+应激组。荷瘤+应激组给予束缚应激,将应激裸鼠放入自制束缚筒中,不挤压裸鼠尾巴,裸鼠可前后活动;单纯荷瘤组正常喂养,不做处理。采用ELISA法检测两组裸鼠血清肾上腺素的含量,免疫组织化学方法检测两组裸鼠移植瘤组织细胞外基质金属蛋白酶诱导因子(EMMPRIN)、细胞增殖相关抗原(Ki-67)、血管内皮生长因子(VEGF)、微血管密度(MVD)和内皮细胞增殖相关抗原(CD105)表达。结果荷瘤+应激组裸鼠移植瘤质量、瘤体积和血清肾上腺素含量均高于单纯荷瘤组,差异有显著性(t=-2.812~-2.486,P<0.05);荷瘤+应激组裸鼠移植瘤组织中EMMPRIN、Ki-67和VEGF的阳性表达水平均明显高于单纯荷瘤组,差异有显著意义(Z=-2.636~-2.033,P<0.05);荷瘤+应激组裸鼠MVD与单纯荷瘤组比较,差异具有显著意义(t=-2.846,P<0.05)。裸鼠移植瘤组织中EMMPRIN与VEGF、Ki-67的表达水平呈正相关(r=0.702、0.534,P<0.05)。结论应激可能通过上调EMMPRIN的表达,诱导癌细胞增殖、侵袭以及血管生成,促进裸鼠胰腺癌的生长。 Objective To create a restraint stress model of transplanted pancreatic cancer (TPC) in nude mice and ex- plore the stress in promoting the growth of the cancer. Methods Twenty mice were evenly randomized to control group (pure- cancer-bearing) and stress group (cancer-bearing-plus-stress group). The mice in the stress group were put in a self-made bucket allowing them to move forward and backward, without squeezing their tails. Those in the control group were fed normally. ELISA was used to detect serum adrenalin; the expressions of extracellular matrix metalloproteinase inducer (EMMPRIN), Ki-67, vascu- lar endothelial growth {actor (VEGF), microvessel density (MVD) and CD105 in tissues of transplanted tumors were detected by immunohistochemintry in both groups. Results The weight and volume of transplanted tumors as well as serum adrenal levels in the stress group were higher than that in the control (t =- 2.812-- 2.486, P d0.05). The positive expressions of EMMPRIN, Ki-67 and VEGF in the stress group were higher than that in the control (Z=--2.fi36-- --2.033 ,P(0.05). The difference of MVD between the stress group and the control was significant (t =-2.846,P〈0.05). The expression of EMMPRIN was positively cor- related with VEGF and Ki-67 in transplanted tumors (r=0.702,0.534;P〈0.05). Conclusion Stress promotes the growth of pancreatic cancer in nude mice is likely through up-regulating the expression of EMMPRIN, and inducing cancer cell proliferation, invasion and angiogenesis.
