摘要
目的:比较从全血中3种提取DNA方法所需要的时间、样本量、提取的DNA纯度、产量以及方法本身的优缺点.方法:采用蛋白酶K法、氯仿-异戊醇法、碘化钾法直接从全血中提取基因组DNA.结果表明:3种方法所提取的DNA质量均能达到分子生物学的实验要求,但碘化钾法提取的DNA纯度最高,且操作时间最短,该法简便、快速和DNA收获率高.
Three methods, protease K, chloroform-isoamyol and potassium iodide methods, for extracting human genomic DNA from whole blood were compared on the time required, sample size, DNA purity, quantification of DNA and their advantages and disadvantages. The qualities of the DNA extracted from all the three methods could meet the requirements of the molecular biological experiments. Potassium iodide method harvested the best quality DNA and required the least time. Potassium iodide method was simple, rapid and efficient, and could be used for routine DNA extraction from whole human blood from the findings.
