摘要
目的:探讨完全睡眠剥夺对大鼠caspase-3的影响及其导致记忆、认知等功能障碍的机制。方法:取Sprague-Dawley大鼠35只,随机分为正常对照组、大平台对照组和完全睡眠剥夺组。其中正常对照组(NC)5只大鼠,正常笼养;大平台对照组(TC)只有72、96及120 h 3个时间点,每个时间点5只大鼠;完全睡眠剥夺组(TSD)也有72、96及120 h 3个时间点,每个时间点5只大鼠。采用小平台水环境法建立完全睡眠剥夺模型,在不同的时点处死大鼠后运用免疫组化SP法检测各组大鼠海马和皮质caspase-3的分布规律和动态变化。结果:caspase-3在各组的海马和皮质均有表达,随着睡眠剥夺时间的延长,caspase-3的表达逐渐增加,与正常对照组、大平台对照组差异有显著性(P<0.01)。结论:完全睡眠剥夺能引起大鼠海马和皮质caspase-3的表达增加,并随睡眠剥夺时间的延长而渐趋明显。可能是完全睡眠剥夺引起大鼠神经元调亡,进而引起大鼠记忆、认知等功能障碍的机制之一。
Objective:To investigate the effect of total sleep deprivation on expression of caspase-3 in rat crotex and hippocampus. Methods:Twenty -five adult male Sprague-Dawley rats were divided randomly into normal control group(n=5),tank control group (n=5) and total sleep deprivation group(n=15). The animals in TSD group were sacrificed at the following time-points 72 h ,96 h and 120 h after TSD,respectively.By strept avidin immunostaining,the expression and distilbution of caspase-3 were detected in the cortex and hippoeampus.Results:After TSD 72h, the expression of caspase-3 in TSD group in rat cortex and hippocampus was higher than that in nor- real control group and tank control group (P〈0.01),with prolonged deprivation time,the numbers of immuno-positive cell increased obvously.Later time point subgroup was compared with the former subgroup,with evident statistics significance (P〈0.01).Conclusion:Total sleep deprivation might increase the expression of caspase-3 in rat cortex and hippoeampus. The result suggests that caspase-3 might be one of neurobiological alterations leading to brain dysfunction such as learn and memory disturbance.
