摘要 目的:探索大鼠不同时间点睡眠剥夺后脑干中缝核群星形胶质细胞(astrocyte,AS)的反应。方法:采用小平台水环境法建立大鼠睡眠剥夺模型,用免疫组化的方法测量胶原纤维酸性蛋白(glialfibrillaryacidicprotein,GFAP)在脑干中缝核群的表达,分组为睡眠剥夺3,6,12,24,96h组,大平台对照组和正常单独饲养组,每组4只。结果:睡眠剥夺后GFAP在中缝核群均有表达,中缝苍白核和中缝背核有较高表达,随着睡眠剥夺时间的延长,中缝苍白核表达逐渐减少,中缝背核表达逐渐增加。结论:睡眠剥夺影响GFAP蛋白的表达,AS参与睡眠调节。 AIM:To investigate the reaction of astrocytes of the rapheal nuclei in brain stem following sleep deprivation at different time points.METHODS:A total of 42 Sprague-Dawley rats were used to establish sleep deprivation models on small platforms over water,and then divided into 3,6,12,24 and 96-hour sleep deprivation and large platform groups of 4 cases in each group.The left two rats were used as normal controls feeding individually.Immunohistochemistry technique was performed to detect the expression of glial fibrillary acidic protein(GFAP) in the rapheal nuclei of brain stem.RESULTS:The expression of GFAP was found in all the groups,especially higher in nucleus raphes pallidus and nucleus raphes dorsalis.With the time,the expression of nucleus raphes dorsalis increased gradually,while that of nucleus raphes pallidus decreased.CONCLUSION:Sleep deprivation has an effect on GFAP protein expression,and astrocytes are involved in the regulation of sleeping.
