摘要
目的:探讨白芍总甙(TGP)对类风湿关节炎(RA)患者外周血单个核细胞(PBMC)产生趋化因子配体3(CCL3)、趋化因子配体5(CCL5)的影响。方法:采用酶联免疫吸附法(ELISA)测定20例RA患者血清中的CCL3、CCL5水平,并与健康对照组比较。分离20例RA患者的PBMC进行体外培养,脂多糖(LPS)诱导CCL3、CCL5产生后,加入不同浓度的TGP,采用酶联免疫吸附法(ELISA)测定培养上清中的CCL3、CCL5水平。结果:RA患者血清中CCL3、CCL5水平均高于健康对照组。TGP浓度达到5μg/mL时可显著性抑制CCL3产生,TGP浓度达20μg/mL时可显著性抑制CCL5的产生,在5~40μg/mL浓度范围内呈剂量依赖性,之后继续增大剂量,其抑制作用亦不再增强。结论:TGP能够抑制RA患者PBMC中趋化因子的产生,这可能是其治疗类风湿关节炎的机制之一。
Objective To explore the inhibitory effects of total glucosides of paeony, on chemokine CCL3 and CCL5 production in Peripheral Blood Mononuclear Cells (PBMC) isolated from patients with rheumatoid arthritis. Methods The levels of chemokine CCL3 and CCL5 in serum of 20 RA patients were determined by enzyme -linked immunosorbent assay (ELISA) , then compared with the healthy controls. PBMC isolated from 20 RA patients were cultured and stimulated with lipopolysaccharide (LPS) , then given different dosage of TGP respectively. The levels of CCL3 and CCL5 in cultural supematants were also determined by ELISA. Results The serum levels of CCL3 and CCL5 in RA patients were significantly higher than healthy controls, TGP was found to significantly inhibit LPS -induced CCL3 production at the concentration of 5μg/mL, and significantly inhibit LPS- induced CCL5 production at the concentration of 20μg/mL in RA PBMC. Moreover, the inhibitory effect of TGP were in a dose-dependent manner in the concentration ranged 5 - 40 μg/mL. CONClUSiON TGP can inhibit chemokine production so as to alleviate the inflammation of rheumatoid arthritis.
