不同浓度的IL-1β和TGF-β_1对大鼠肋软骨细胞的影响 被引量:2

The effects of IL-1β and TGF-β_1 on rat costochondral chondrocytes

摘要 目的:研究人白细胞介素-1β(IL-1β)和转化生长因子-β1(TGF-β1)对体外培养的SD大鼠肋软骨细胞功能的影响。方法:采用免疫组织化学检测不同剂量IL-1β和TGF-β1作用48h后软骨细胞Ⅱ型胶原和蛋白多糖表达水平。采用RT-PCR方法检测细胞Ⅱ型胶原、aggrecanase-1,2、aggrecan的mRNA的表达水平。结果:IL-1β可以促进软骨细胞aggrecanase-1,2的表达,从而降低多聚蛋白聚糖的含量,破环细胞外基质结构,减少Ⅱ型胶原的含量,使细胞呈现去分化的表现,对软骨细胞起负性调节作用。低浓度的TGF-β1对软骨细胞起保护的作用;高浓度的TGF-β(1100ng/ml)在促进蛋白多糖表达的同时也促进软骨细胞蛋白多糖的降解,从而打破软骨的代谢平衡,起负性调节的作用。 Objective In order to study IL-1β and TGF-β1 have what effects on SD rat chondrocyte cell. Methonds Different dosis of IL-1β and TGF-β1 was used to deal chondrocyte cell 48 hours. Then aggrecan and collagen Ⅱ of those celluar and the control group were detected by immune-histochemistry. RT-PCR methondwas used to assessment celluar aggrecan and collagen Ⅱ and aggrecanase-1,2 mRNA expressing. Results IL-1β increases the expressing of aggrecanase-1,2 accordingly decrease the content of aggrecan, reduce the celluar character collagen Ⅱ, destroy the integrity of ECM. IL-1β have negative effects on chondrocytes. Low-grade TGF-β1 have energetic effects on chondrocytes, altus improve the content of aggrecan also degration the aggrecan ,finally breakdown the balance of ECM, have negative effects on chondrocytes.
