摘要
目的筛选制备猴头菇浸提液的最优工艺条件及研究猴头菇多糖的免疫增强作用。方法将猴头菇粉碎、脱脂,然后经水浸提液浸提,再经减压浓缩、Sevag法去除游离蛋白、乙醇沉淀得到猴头菇多糖。采用3H-TDR法测定小鼠LAK细胞活性。结果经正交实验得到制备浸提液的最优工艺条件:温度85℃,时间4 h,加水量150 ml;除蛋白的最优工艺条件:样液:氯仿-正丁醇(体积比)=3;当乙醇体积分数为75%时可沉淀出较多的糖。给予猴头菇多糖小鼠LAK细胞抗肿瘤活性增强。结论利用本工艺可较好的提取猴头菇多糖,且该多糖有显著的免疫活性。
Aim To explore the best method of extracting polysaccharide from Hericium erinaceus and to study its immune-enhancing activity.Methods The extracted polysaccharide is gained by degrease,lixiviation,concentrating,sevag reagent′s getting rid of free protein and precipitation through ethanol.The activity of mice′s LAK cell was determined by ^3H-TDR.Result The orthogonal experiment shows that the best process is temperature at 85℃,time being 4 h,water capacity of 150 ml.The volume ratio of polysaccharide solution to sevag reagent is 3.When the alcohol consistency comes up to 75% in the polysaccharide solution,the polysaccharide is precipitated out more.The Polysaccharide also has a role in activation of mice LAK and IL-2 cell,which could enhance the immune activity.Conclusion Polysaccharide from Hericium erinaceus can be extracted by this method.And the polysaccharide has a remarkable immune-enhanceing activity.
