疏血通对局灶性脑缺血大鼠神经保护和p53表达的影响 被引量:6

Neuroprotective Effects of Shuxuetong Injection on Expression of p53 in Rats with Focal Cerebral Ischemia-Reperfusion Injury

摘要 目的探讨疏血通对大鼠脑缺血再灌注后可能的神经保护作用机制。方法制备脑缺血再灌注模型,用疏血通进行干预,观察脑组织细胞的形态学改变,p53蛋白表达,细胞原位凋亡情况,脑梗死体积的变化。结果大鼠脑缺血2h再灌注24h,治疗组的脑梗死体积较缺血组明显缩小(P<0.01)。治疗组大鼠的神经功能缺损评分,在6h时间点与缺血组比较无统计学意义,其余时间点均明显低于缺血组(P<0.05或P<0.01)。缺血组和治疗组缺血再灌注6h均可见凋亡细胞,表达高峰分别为48h、72h,治疗组的表达高峰时间延迟,且数量明显减少(P<0.05)。p53蛋白在缺血再灌注后6h出现表达,缺血组的表达高峰为48h,治疗组的表达高峰为72h,治疗组各时间点p53阳性细胞数较缺血组明显减少(P<0.05或P<0.01)。结论疏血通可减轻大鼠脑缺血再灌注损伤,缩小梗死体积,抑制p53蛋白的表达,改善神经功能。 Objective To explore neuroprotective effects of Shuxuetong injection on rat brain after cerebral ischemia - reperfusion injury. Methods Nine - six SD rats were divided into control group (n=6), sham operation group (n=6), ischemic group (n=42) and treatment group (n =42). The models of ischemia- reperfusion injury were established and morphology changes, expression of p53 protein, apoptosis and infarction volume were studied. Infarction volume was tested by TTC staining. Results Infarction volume in Shuxuetong injection group was smaller than that in control group (P〈0. 01). The neural function in Shuxuetong injection group was better than that in control group at all the time except at 6 h, TUNEL - positive cells were more apparent in the perifocal tissue and particularly in the inner border regions immediately adjacent to the ischemic core. Apoptosis cells were found in ischemia group and Shuxuetong group since 6 h after ischemia- reperfusion, and reached peaks at 48 h/72 h after reperfusion respectively. The expression peak was delayed and the amount of apoptosis cells were decreased remarkably in the Shuxuetong group (P(0.05). The expression of p53 protein were found at 6h after ischemia- reperfusion, and reached peak at 48 h time point in the ischemia group, and it reached peak at 72h time point in the treatment group. The amount of immunity - positive cells of p53 were less obviously than that in ischemia group (P(0.05). Conclusion The result suggested that Shuxuetong injection could significantly reduce the cerebral ischemia- reperfusion injury, decrease the infarction volume and improve neural function. The neuroprotction against cerebral ischemia reperfusion injury might be related to down - regulation of p53 and to inhibit apoptosis.
