表达金丝桃素合成酶的基因工程菌发酵工艺研究 被引量:1

摘要 采用摇瓶发酵试验,分别对影响工程菌生长和表达的条件:培养基初始pH值、诱导温度、诱导时间、诱导物浓度等进行了考察,优化了重组大肠杆菌的发酵条件,确定了周期短、产率高且稳定可靠的发酵工艺,即工程菌在pH值为7.4、温度为25℃、IPTG(异丙基硫代-D-半乳糖)浓度为0.8mmol/L时蛋白表达量最高。 To develop the optimal fermentation conditions of genetic recombinant Escherichia coll. The expression of target protein was analyzed to optimize the range of pH values, induction time and induction temperature in the process of fermentation. The established fermentative procedure increased the expression and collection efficiency of recombinant protein.
