摘要
在低离子强度下,meso[四(对三甲铵基)苯基]卟啉(TAPP)能在核酸分子表面进行长距组装并诱导核酸超螺旋结构的形成.长距组装使得TAPP的Soret带发生减色效应,并使Soret带处激发产生的荧光被猝灭.体系离子强度(I)增大将降低TAPP长距组装的能力,导致荧光猝灭程度降低.在I=0004时,荧光猝灭程度与核酸的浓度成线性关系,可用于15×10-6~90×10-6mol/L的小牛胸腺DNA,15×10-6~105×10-6mol/L鱼精子DNA和酵母RNA的测定.
At low ionic strength,meso tetrakis[4 (trimethyammonium)phenyl]porphine(TAPP) can induce the formation of suprahelical helixes of nucleic acids because of its long range assembly on the surface of nucleic acid molecules,which leads to the hypochromic effect of the Soret band of TAPP and quenching of the fluorescence excited at the Soret maxima.The fluorescence quenching effect was decreased with of increasing ionic strength of the system.At ionic strength 0 004,linear fluorescence quenching was found in the range 1 5×10 -6 ~9 0×10 -6 mol/L of calf thymus DNA,1 5×10 -6 ~10 0×10 -6 mol/L of fish sperm DNA and yeast RNA.The determination results for 6 synthetic samples were satisfactory.
