A STUDY ON CYTOKINESIS BLOCK MICRONULEUS (CBMN) TECHNOLOGY IN CHO CELL

摘要 使用胞质分裂阻断微核(CBMN)法,采用CHO细胞评价一些环境因子造成的染色体损伤和细胞毒性.当细胞松弛素B(CB)的剂量为6mg·L-1,作用时间为40h时,就能获得一个含有适宜数量双核细胞和多核细胞的CHO细胞体系.利用这一体系,研究了一种标准的实验室致突变剂丝裂霉素C(MMC)对CHO细胞的影响,在实验剂量范围内,得到了可重复诱导微核形成的效果,并与其他实验室的研究结果一致.此外还检测了另外2项毒性指数:核分裂指数(NDI)和胞质分裂阻断增殖指数(CBPI).为采用多参数分析检测一些污染物质导致的遗传毒性提供了一种很有价值的方法. This work is aimed to develop and use the cytokinesis block micronucleus (CBMN) method in Chinese hamster ovary (CHO) cell in order to evaluate the chromosome damage and cytotoxicity induced by some environmental factors. With 6mg·L-1 cytochalasin B(CB) treated for 40h, a CHO cell system including optimum numbers of binucleated and multinucleated cells can be obtained. By using the cell system. the effects on CHO cell of a standard laboratory mutagen mitomycin C (MMC) were tested, a positive and repetitive results of the micronucleus formation were obtained. These resulte are in agreement with other studies.Additionally, by means of the cell system, two cytotoxicity indexs: nuclear division index (NDI)and cytokinesis block proliferation index (CBPI) were also used to assess. By using the CBMN method in CHO, the valuable multiparameter analysis can be used for detecting the genotoxicity induced by some pollutants.
