摘要
观察了3种已报道的脱色液在MTT比色法中的使用效果,实验证明它们均存在一定的局限性,不适于微囊细胞的脱色。改良的脱色液配方为N,N-二甲基甲酰胺-Triton-水(DTW),它不仅可迅速完成脱色,呈色稳定,不引起蛋白质沉淀,且无需破囊和洗涤细胞,从而较好地解决了微囊固定化细胞计数的难题。
Based on Mosmann's MTT method, a modified MTT system was developed and
applied to the measurement of cell growth in microcapsules. In comparison with three kinds of
pubilshed extraction buffer, a new lysing buffer N, N-dimethylformamide-Triton-water (DTW)
showed a lot of advantages. It could extract formazan dye out of microcapsules completely and
quickly, the OD value was stable during 24 hours, and it was unnecessary to break the capsules, to
centrifuge and wash the cells. So, it is very suitable for counting the cells cultivated in
microcapsules.
