摘要 利用化学发光分析技术建立一种新的腺苷脱氨酶活性测定方法,该方法简便易行,快速灵敏。酶反应的最佳条件:底物 Km 为0.31mmol/L,最佳 pH 为7.40,反应时间为10分钟。该方法的重复性CV 为2.74%,该方法测量结果与比色法测量结果有很好的相关性(r=0.986,P<0.01)。将该方法应用于结核性胸膜炎和癌性胸水的鉴别诊断,结果表明,癌性胸水的 ADA 活性显著低于结核性胸水(P<0.01)。由此可见,用化学发光法测定腺苷脱氨酶活性不仅精确度高,而且检便易行,对结核性胸膜炎和癌性胸水的鉴别诊断有较高的实用价值。 A novel analytical method of adenosine deaminase(ADA)in body fluids by luminescent assay was described in this paper.This method is simple,rapid,sensitive and accurate.Optimum reaction condi- tions for activity of ADA were investigated.The substrate Km was 0.31mmol/L.The optimum pH was 7.4, The reaction time was 10 minutes:The precision of this method is 2.74% and results from luminescent assay is significantly related to results from colormetrie assay(r=0.986,P<0.01).ADA of patients with tubercu- lous pleuritis is more active than that of patients with cancerous pleuritis.The luminescent method can be ef- fectively applied in discrimination between tuberculous pleuritis and cancerous pleuritis.
