摘要
目的:观察β-catenin在毛囊干细胞中的分布表达变化,探讨其与细胞增殖的关系。方法:体外分离培养毛囊干细胞,绘制细胞生长曲线,采用ICC方法观察β-catenin在不同培养时相毛囊干细胞中的定位表达变化。结果:培养2d便可见细胞生长,6d时细胞数达到最高峰;培养前3d β-catenin表达于胞浆及胞膜,第5—6天出现核内表达,伴有ki67核内表达;继续培养,β-catenin集中在胞核表达,胞浆呈弱表达,并表现与细胞增殖的相关性。结论:β-catenin核内转位是毛囊干细胞增殖的重要信号调节机制。
Objective:To investigate the translocated expression of β -catenin in hair follicle stem cells (HF- SCs) in vitro, and to further explore the relationship between β -catenin and cell proliferation. Methods: HFSCs were obtained from rat follicles by microdissection. Following the growth curve definited, the immunocytochemistry was used to detect the expression of β -catenin in different phases. Results: The growth curve showed that cells proliferated quickly at day 3 after inoculation, and the cell proliferation reached the peak at the day 6 and main- tained in the following days. Meanwhile, β - catenin was obviously expressed in cytoplasm and cell membrane of HFSCs in the third day. After 5 to 6 days of culture, β - catenin appeared in nuclear, accompanied with strong expression of ki67, a nuclear proliferative antigen. Then, β - catenin was translocated in nuclear completely instead of cytoplasm. Conclusion : All the data displayed a correlation between β - catenin and cell proliferation, suggesting that β -catenin might be an essential regulative molecule involving in HFSC proliferation.
