摘要
目的观察铀矿粉对体外培养的支气管上皮细胞的毒性作用,以进一步了解其引起肺癌的可能机制。方法将不同浓度的铀矿粉与体外培养的支气管上皮细胞进行长时间接触,分为对照组、染毒组、纳米铀矿粉组和二甲亚砜保护组,通过血清抗性实验、锚着独立生长实验等,观察不同组间细胞受损的情况。结果随着铀矿粉浓度的增高,支气管上皮细胞受损明显加重,加入二甲亚砜的支气管上皮细胞受损情况好于未加入者。但纳米级铀矿粉对支气管上皮细胞的损伤且介于二者之间。结论铀矿粉对支气管上皮细胞有明显的恶性转化作用,可能是铀矿粉对肺损伤的主要机制,二甲亚砜具有一定的保护作用。而纳米级的铀矿粉作用机制可能有别于普通铀矿粉,需进一步研究。
[ Objective]The aim of this study was to explore the malignant transformation effect on cytotoxic of human bronchial epithelial cell (BEAS-2B) induced by uranium mine dust, further investigate the cause and mechanism of human lung cancer. [Methods] The immortalized human bronchial epithelial ceils BEAS-2B were treated with uranium mine dust at the concentrations of different dose,including a negative control group and the group protected by DMSO. The characteristics of the cellular biology and the malignant transformation phenotype of cells were identified through observing serum resistance, anchorage independent growth, et al. [Results] The mutation frequency of BEAS-2B cells were significantly increased by uranium mine dust and the damaged degree will aggravate with high concentration. Uranium mine dust induced cell damage will reduced effectively by protected of DMSO. But the damaged degree of the cells reduced by Nano group is between the uranium mine dust group and the group protected of DMSO. [Conclusons] Uranium mine dust could induce malignant transformation of BEAS-2B cells.DMSO can prevent damage of Uranium mine dust. But the mechanism of Nano Uranium mine dust will be studied in the future.
