摘要 目的:以生物医学工程的角度,研究基于数字图像处理技术的鸡胚绒毛尿囊膜图像血管自动分析方法,设计和研制专用实验检测系统。方法:实验于2005-06/2007-06分别在上海理工大学医疗器械与食品学院中心实验室和上海中医药大学进行。根据鸡胚绒毛尿囊膜血管图像的特点,研究和开发了包括鸡胚绒毛尿囊膜血管数字图像背景褪色、血管增强、背景和血管定标、标尺校准、血管测量区域划分、血管面积及面积比参数测量等主要分析软件,以及相应的测量分析系统。通过实验检验分析方法和测量参数,并对分析系统进行改进。结果:建立了有效的鸡胚绒毛尿囊膜数字图像血管自动分析的方法和步骤,研制了专用的实验分析系统。通过药物实验对鸡胚绒毛尿囊膜血管图像进行了参数检测,获取了多组实验数据,并进行了统计分析。统计分析结果显示试验药物有抑制血管生长作用,且作用大小呈剂量依赖关系。结论:经实验证明基于数字图像处理技术的鸡胚绒毛尿囊膜图像血管自动分析方法能提供可靠的鸡胚绒毛尿囊膜模型定量分析数据,并且在分析速度、测量精度、数据重复性等方面均优于常规人工目测血管计数方法。 AIM: To study the method for automatic analysis of blood vessels in digital chick embryo chorioallantoic membrane (CAM) images and develop a special experimental analysis system based on digital image processing and biomedical engineering. METHODS: The experiment was performed at the Central Laboratory of School of Medical Instrument and Food Engineering, University of Shanghai for Science and Technology, and Shanghai University of Traditional Chinese Medicine from June 2005 to June 2007. Based on the characteristics of CAM images, the image analysis software and hardware were studied and developed. The main functions of the analysis software included CAM image background fading, blood vessels enhancement, background and blood vessel calibration, measurement region division, micro-ruler calibration, blood vessel area and area ratio measurement and so on. The analysis method and measurement parameters were tested by experiments, and the analysis system was improved during that process. RESULTS: The analysis method and procedure as well as the analysis system for CAM image were efficiently established. The parameter measurements in CAM images from drug experiments in which the animals were divided into several groups were executed. The data obtained from the experiments were statistically analyzed. The result of statistical analysis indicated that the tested drug could restrain the growth of blood vessels in a dose-dependent manner. CONCLUSION: The method and the system are reliable for the analysis and can provide quantitative data from CAM model. It is also better than conventional method of blood vessel count from CAM images by microscope in analysis speed, precision and data reproducibility.
