摘要
目的探讨左归丸、右归丸对实验性自身免疫性脑脊髓炎(EAE)大鼠病程及其血浆白细胞介素(IL-1α、IL-4)、干扰素-γ(IFN-γ)和肿瘤坏死因子-α(TNF-α)的影响。方法用髓鞘碱性蛋白(MBP)免疫Lewis大鼠诱导EAE模型,随机分为模型组、激素组、左归丸组、右归丸组及正常对照组,研究各组间发病率、死亡率、潜伏期、病程、体重及神经功能评分的差异,并分别在急性期(免疫后第15天)和缓解期(免疫后第27天)采集血样,检测血浆细胞因子水平。结果右归丸组发病率低于其他各处理组(P<0.05);急性期和缓解期,右归丸组体重减轻程度最轻,体重均高于激素组和左归丸组(P<0.05);急性期神经功能评分左归丸组和右归丸组均低于模型组和激素组(P<0.05);急性期左归丸组血浆IL-4水平高于正常对照组和激素组,IFN-γ和TNF-α水平高于正常对照组、模型组和激素组(P<0.05);缓解期右归丸组血浆IL-4和TNF-α水平高于正常对照组和模型组,IFN-γ水平高于正常对照组(P<0.05);缓解期模型组血浆IL-1α水平高于正常对照组(P<0.05);模型组缓解期血浆IL-1α水平高于急性期(P<0.05);左归丸组缓解期血浆IL-4、IFN-γ、TNF-α水平低于急性期(P<0.05)。结论右归丸可降低EAE发病率;左归丸和右归丸均能减轻EAE疾病严重程度,治疗效果优于醋酸泼尼松;右归丸具有减轻EAE大鼠体重下降的作用;IL-1α在EAE疾病过程中发挥重要作用,醋酸泼尼松、左归丸和右归丸可下调EAE缓解期血浆IL-1α水平;左归丸可上调EAE急性期血浆IFN-γ、TNF-α、IL-4水平;右归丸可上调EAE缓解期血浆IFN-γ、TNF-α、IL-4水平。
Objective To study the effect of Zuogui Pills and Yougui Pills on clinical course and the levels of interleukin-1 α ( IL-1α), interleukin-4 ( IL-4 ), interferon-γ (IFN-γ) and tumor necrosis factor- α(TNF-α) in plasma of experimental autoimmune encephalomyelitis (EAE) Lewis rats. Methods Lewis rats were immunized with the myelin basic protein (MBP). Rats were divided randomly into normal-group (Group A) EAE-group (Group B), prednisone-group (Group C), Zuogui Pills group ( Group D) and Yougui pills group ( Group E) after immunization. Rats were monitored daily, the weight and the clinical score of the degree of paralysis were recorded. On day 15 and 27 after immunization, rats were killed and the peripheral blood was collected for cytokines examination. Results The incidence of EAE was significantly lower ( P 〈 0.05 ) and the weight was significantly heavier ( P 〈 0.05 ) in Group E than in other groups, in Group E. The paralysis score was significantly lower ( P 〈 0.05 ) in Group D and Group E than in Group B and Group C. On day 15 after immunization, the level of IL-4 was significantly higherin Group D than in Group A and Group C ( P 〈 0.05 ), and the level of IFN-γ and TNF-α were significantly higher in Group D than in Group A, Group B and Group C ( P 〈 0.05 ). On day 27 after immunization, the level of IL-1α was significantly higher in Group B than in Group A ( P 〈 0.05 ), the level of IL-4 and TNF-α was significantly higher in Group E than in Group A and Group B ( P 〈 0.05 ), and the level of IFN-γ was significantly higher in Group E than in Group A(P 〈0.05). The level of IL- 1α of Group B was significantly higher on day 15 than on day 27 after immunization (P 〈0.05). The level of IL-4, IFN-γ and TNF-α of Group D was significantly higher on day 15 than on day 27 after immunization (P 〈0.05). Conclusion Yougni Pills can decrease the incidence of EAE. Zuogni Pills and Yougni Pills both are better than prednisone in alleviating clinical signs. Yougni Pills has the effect on increasing weight of EAE rats. Prednisone, Zuogni Pills and Yougni Pills can down-regulate the level of IL-1α in serum. Zuogni Pills can up-regulate the level of IFN-γ, TNF-α and IL-4 in acute phase. Yougni Pills can up-regulate the level of IFN-γ, TNF-α and IL-4 in paracmasis.
