摘要
目的探讨基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)在多发性硬化(MS)动物模型—实验性自身免疫性脑脊髓炎(experimental autoimmune encephalomyelitis,EAE)大鼠脑脊髓中的表达和作用。方法应用豚鼠脑脊髓匀浆与完全福(氏)佐剂(CFA)混匀于Wistar大鼠颈背部皮下注射诱导建立EAE模型,同时将单独注射CFA或NS的大鼠设为对照组。采用半定量免疫组化方法检测Wistar大鼠发病后72 h内及71、4 d不同时间段脑脊髓中MMP-9的表达,并与对照组比较。结果经免疫组化方法检测结果显示EAE大鼠脑脊髓内有特异的MMP-9表达,与对照组比较差异有显著性。发病72h内MMP-9表达高于发病后7d(P<0.05)及发病后14 d(P<0.01)。结论EAE大鼠脑脊髓的单核淋巴样细胞有特异MMP-9表达,且发病早期MMP-9阳性细胞率高于发病后期、恢复期及对照组,提示MMP-9与EAE发病有关。
Objective To investigate the expression and functions of matrix metalloproteinase-9 (MMP-9)in the Wistar rat of experimental autoimmune encephalomyelitis(EAE). Methods EAE model was made by subcutaneous injection at the nape with guinea pig brain-spinal cord homogenate in CFA. The control model was injected subcutaneously with CFA or NS respectively. The expression of MMP -9 was detected by immunohistochemistry in brain and spinal cord of EAE rat on ≤72 h, days 7 and days 14 after clinical syndrome onset, the result was compared with that of control model. Results Immunohistochemistry showed specific MMP-9 expression in EAE brain and spinal cord by contrast to control model. The expression of MMP-9 in EAE model on ≤72 h had a significant enhancement as compared to that on days 7 (P〈0.05)and days 14 (P〈0.01). Conclusions Specific MMP -9 expression was found by immunohistochemistry in mononuclear ceils in EAE brain and spinal cord. The percentage of MMP-9 positive mononuclear cells in EAE at early stage of onset was higher than that of the later stage of onset, and abated obviously at the stage of recovery which prompts that MMP-9 might participate in the etiology of EAE.
