摘要
目的建立适合基层医院普及的茶碱血药浓度测定方法。方法氯仿/异丙醇混合溶媒提取,碱液反提取,调节pH值避免巴比妥类药物干扰,应用紫外分光光度法测定茶碱浓度。结果茶碱浓度在5～40mg/L之间,标准曲线方程是C=55.8659A-0.0112(r=0.9992,n=6)。平均回收率是98.61%(RSD=2.09%,n=5)。变异系数是2.83%。结论本方法所需条件简便,测定快速、准确,适合基层普及。
Objective To develop a method for determination of the theophylline in the blood,which is worth popularizing in the basic-hospital. Methods The determination of theophylline by ultraviolet spectrophotometry was based on the extraction with the mixture solvent of 2-propanol with chloroform,back extraction with alkali solution and pH adjusting for elimination of the interference of barbital. Results In the range of 5 rag/L- 40 mg/L for theophylline, the equation of calibration curve is C = 55. 8659A-0.0112,with the correlation coefficient r = 0. 9992 （ n = 6 ）. The average recovery is 98.61% （ RSD = 2.09%, n = 5 ）. The coefficient of variation （CV） is 2.83%. Conclusion The method is simple, rapid, accurate and suitable for the popularization in the basic-hospital.
