刺五加多糖对H_2O_2诱导海马细胞凋亡的保护作用 被引量:1

摘要 目的研究刺五加多糖(Acanthopanax senticosus polysaccharide,ASPS)对大鼠海马细胞的抗凋亡作用,并探讨其机制。方法运用海马神经细胞原代培养技术,采用H2O2诱导建立细胞凋亡模型。观察细胞形态学变化;MTT法测定细胞活性;免疫组化法检测细胞中Bax与Bcl-2的蛋白表达。结果形态学观察显示:与模型组比较,ASPS组细胞损伤程度明显减轻;ASPS组细胞活性(0.46)比模型组细胞活性(0.36)高(P<0.01),ASPS组Bcl-2表达量较高而Bax表达量较低。结论ASPS能够提高海马神经细胞的抗凋亡能力,并具有一定的浓度依赖性。 Objective To investigate the effect of acanthopanacis senticosi polysaccharides (ASPS) on apoptosis of rat hippocampal neurons. Methods The hippocampal neurons were isolated from SD rats aged less than 3 days and cultured, then induced to apoptosis by H202. After treated with ASPS, the changes of cell form were observed, the viability of hippocampal neurons was detected with MTT method and the expressions of Bax and Bcl-2 were detected by immuno- histochemical method. Results The ceil injury of hippocampal neurons in ASPS-treated group was slighter than that in model group. The viability of hippocampal neurons in ASPS-treated group (0. 46) was higher than that in model group (0. 36) (P〈0. 01). The hippocampal neurons in ASPS-treated group had higher expression of Bcl-2 and lower expression of Bax. Conclusion ASPS enhances the anti-apoptotic capacity of hippocampal neurons in a concentration-dependent manner.
