摘要
目的:研究生地对原代分离的大鼠肺间质成纤维细胞(fibrob last,Fb)Ⅰ、Ⅲ型胶原(ColⅠ、ColⅢ)表达的影响。方法:生地经提取分离后得到不同部位的提取物。大鼠肺间质成纤维细胞经原代分离传代后,加入不同浓度的生地提取物及生地注射液作用72 h,采用RT-PCR的方法测定其中Ⅰ、Ⅲ型胶原的量。结果:水提醇沉上清液中以低聚糖为主的部分除个别浓度外,其余浓度对ColⅠ表达均有抑制作用,高浓度对ColⅢ表达有一定的抑制作用;醇沉沉淀部分2×10-2g/mL的浓度对ColⅠ表达抑制作用明显,各浓度对ColⅢ表达均有抑制作用;生地注射液各浓度均对ColⅠ的表达均有抑制作用,低浓度(5×10-3g/mL)对ColⅠ、ColⅢ表达均有明显抑制作用。结论:生地中某些成分能够抑制大鼠肺间质成纤维细胞ColⅠ、ColⅢ的表达,是生地对肺纤维化起治疗作用的机制之一。
AIM: To investigate the effeet of Rehmannia glutinosa Liboseh on collagen Ⅰ and Ⅲ of in pulmonary interstitial fibroblast in rat. METHODS: The eomponents of Radix Rehmanniae were extraeted and isolated to several parts. Primary pulmonary fibroblasts were separated from lung interstitial tissue of rat. After being repro- dueed for several generations, fibroblasts were eultured in the extraets of Radix Rehmanniae and Shengdi Solution for injeetion in several eoneentrations respeetively for 72 hours, and then eontents of Col Ⅰ and Col Ⅲ of these fibroblast were determined by RT-PCR. RESULTS: The part eontaining mainly oligosaeeharide from preparation that deeoeted with water and deposited with aleohol eould inhibit Col Ⅰ with a few exeeptions eoneentration while it could inhibit Col Ⅲ at high concentration. The part of deposit inhibited Col I obviously in the concentration of 2 - 10-2 g/mL and eould inhibit Col Ⅲ in all the eoneentrations. Shengdi Solution for injeetion eould inhibit Col Ⅲ in all the eoneentrations. CONCLUSION: Some eonstitutents of Radix Rehmanniae earl inhibit the expression of Col Ⅰ and Col Ⅲ of pulmonary fibroblast which is one of mechanisms that Radix Rehmanniae takes effect on fibrosis disease in pulmonary interstitial tissue.
