摘要
目的研究昆布多糖(Thallus laminariae polysaccharides,TLP)对大鼠减肥、降血脂的作用及其机制。方法制作肥胖大鼠模型,随机分成六组,分别以蒸馏水、TLP(100,200,300mg·kg-1·d-1)、奥利司他(orlistat,赛尼可,60mg·kg-1·d-1)和洛伐他汀(lovastatin,10mg·kg-1·d-1)灌胃,为期40d,在实验过程中,按时检测相关指标。结果TLP能明显降低肥胖大鼠的体重、减少大鼠腹腔、肾、生殖器周围脂肪、降低Lee’s指数和肝、肾重量(P<0.05),而且其作用与减肥药赛尼可在统计学上无显著性差异(P>0.05)。TLP能明显降低肥胖大鼠血清三酰甘油(TG)、胆固醇(TC)、提高高密度脂蛋白胆固醇(HDL-C)水平(P<0.05),而在实验期内低密度脂蛋白胆固醇(LDL-C)变化不明显(P>0.05),而且其作用与降脂药洛伐他汀比较无显著性差异(P>0.05)。TLP能明显改善肥胖大鼠血清卵磷脂胆固醇脂酰基转移酶(LCAT)、脂蛋白脂酶(LPL)和胰脂肪酶(PL)活性(P<0.05),而对肝脂肪酶(HL)活性在实验期间影响不明显(P>0.05)。结论TLP对肥胖大鼠具有明显的减肥作用,该作用与常用的减肥药赛尼可比较无显著性差异。TLP同时能降低血清甘油三酯和总胆固醇、改善血清HDL-C水平,而且该作用与降脂药洛伐他汀比较无显著差异。其作用机制可能与增强LCAT、LPL、PL酶活性有关。
OBJECTIVE To study the functions of Thallus laminariae polysaccharides (TLP) and its mechanisms on experimental fatty rats, METHODS Male SD strains rats (50 - 70 g) were randomly divided into the normal group(A) fed normal diet( 10 rats) and model groups(60 rats) fed high-lipids-diet. After eight weeks, the models were prepared successfully, and they were redivided into six groups : the control group ( Ⅰ ), the low-dosage group ( Ⅱ ), the medium-dosage group ( Ⅲ ), the high-dosage group ( Ⅳ ) , the Orlistat group( Ⅴ ) and the lovastatin group(Ⅳ) ( 10 rats each), Group I was fed with water while Group Ⅱ , Ⅲ, Ⅳ were administrated with TLP at the dosages of 100 mg·kg^-1 ,200 mg·kg^-1 and 300 mg·kg^-1 , Group Ⅴ was treated with the drug of Orllstat at the dosage of 60 mg·kg^-1 and Group VI was treated with the drug of lovastatin at the dosages of 10 mg·kg^-1. The experiment lasted 40 days. During the course, the body weight was tested every 10 days, and the fatty tissue, the weight of liver & kidney and the Lee's Index,the fast blood TG, TC, low-density cholesterol (LDL-C), high-density cholesterol (HDL-C), were tested at the 40^th day. In addition, at the 40^th day the activity of the fast blood LCAT, LPL, PL&HL were also determined. RESULTS The body weight ,the fatty tissue, the weight of liver & kidney and the Lee' s Index ,including TG, TC level of Group Ⅱ,Ⅲ,Ⅳ was decreased significantly compared with that of before administration and Group Ⅰ ( P 〈 0, 05 ). However, there was no difference in Group Ⅱ , Ⅲ, Ⅳ ( P 〉 0.05 ). TLP played an important role in increasing the concentration of blood HDL-C (P 〈 0.05 ). However, there was on significant effect on the concentration of the fast blood LDL-C, TLP could improved the activities of LCAT, PL and LPL significantly ( P 〈 0.05 ). No sig- nificant differences were observed in Group Ⅱ , Ⅲ, Ⅳ( P 〉 0.05 ), However the activity of HL had not changed significantly, CON- CLUSION TLP can significantly lower the body weight and the blood TG, TC levels in hyperlipidemia rats. The function may be related to its improving the activities of LCAT, PL and LPL greatly and increasing the concentration of blood HDL-C.
