摘要
目的探讨RASSF1A与CyclinD1蛋白在乳腺癌发生发展中的作用及其在乳腺癌早期诊断中的意义。方法采用Western blot印迹法检测RASSF1A及CyclinD1在20例乳腺纤维腺瘤组织,20例乳腺导管上皮不典型增生组织,20例乳腺导管内癌组织及50例原发性乳腺浸润性导管癌组织中的表达水平。结果导管内癌组RASSF1A的表达低于不典型增生组(P<0.01),而cyclinD1的表达高于不典型增生组。乳腺癌组织中,RASSF1A的表达与病理分级及临床分期有关(P<0.05)。随着病变程度加重,CyclinD1表达呈增高趋势(P<0.01)。乳腺癌组织中,CyclinD1的表达仅与病理分级有关(P<0.05)。乳腺癌中RASSF1A与CyclinD1两蛋白的表达呈负相关(P<0.05)。结论RASSF1A与CyclinD1两种蛋白的异常表达在乳腺癌的发生发展中可能具有协同作用,二者联合检测能为乳腺癌的早期临床诊断和治疗提供有利的生物学信息。
Objective To investigate the role of the expression of RASSF1A and CyclinD1 in breast cancer and premalignant lesion and its significance in diagnosis of early breast cancer. Methods The expression of RASSF1A and CyclinD1 was detected by Westem blot in 20 cases of breast fibroadenoma, 20 cases of duct atypical intraductal hyperplasia, 20 cases of intraductal breast carcinoma and 50 cases of primarily invasive ductal carcinoma. Results Expression of RASSF1A was lower in intraductal breast carcinoma than in duct atypical intraductal hyperplasia (P〈 0.05 ), but expression of cyclinD1 was higher than in duct atypical intraductal hyperplasia (P〈 0.05). Expression of RASSF1A was associated to TNM stage and pathological grade in breast cancer (P 〈 0.05 ), while expression of CyclinD1 was associated with pathological grade (P 〈 0.05 ). Expression of RASSF1A was negatively correlated with expression of CyclinD1 in breast cancer. Conclusion The silencing of RASSF1A and the expression of CyclinD1 may play an important role in breast cancer pathogenesis.
