摘要
目的通过研究去势前后大鼠下丘脑外侧区(lateral hypothalamic area,LHA)一氧化氮合酶(nitric oxide synthase,NOS)阳性神经元的分布与形态变化,进一步探讨LHA神经细胞参与生殖活动的作用机制。方法将20只实验大鼠分为对照组、去势组、去势给药一月组及去势给药两月组4组,每组5只。各组大鼠脑组织作冰冻切片,行免疫组织化学染色,在光学显微镜下观察LHA的NOS阳性神经元。应用图像分析系统对各切片LHA的NOS阳性神经元进行计数并计算其密度,测定阳性神经元的灰度值。结果去势组LHA的NOS阳性神经元密度值和灰度值都比对照组和去势给药一月组、去势给药两月组高,差异具有统计学意义;而对照组与去势给药一月组、去势给药两月组的阳性神经元的密度以及平均灰度值比较均无统计学差异。结论LHA的NOS可能参与了雌激素对垂体-性腺轴的负反馈调节。
Objective To study the distribution and morphological changes of nitric oxide synthase positive neurons in lateral hypothalamic area(LHA) of female rats before and after ovariectomy, and to explore the effect of neurons in LHA on reproduction. Methods Twenty female rats were divided into 4 groups ( n = 5 in each group) : normal control group, ovariectomized group, 1 month group and 2 month group. The rats in 1 month group and 2 month group were given estrogen replacement therapy for 1 month and 2 months following ovariectary, respectively. Using immunohistochemical method, morphology of NOS positive neurons in LHA were observed under light microscope. NOS positive neurons in LHA were counted and measured by image analysis. Results NOS positive neurons of LHA had higher density in ovariectomy group than in normal control group, estrogen replacement therapy groups (P 〈0.05). Compared with normal control group, NOS positive neurons in LHA had no significant difference in 1 month group and 2 month group. Conclusion The results suggest that NOS may participate in the regulation mechanism of negative feedback between estrogen and hypothalamic-pituitary-gonadal axis.
