Determination of Flavonoid Aglycones in Trachelospermum Jasminoides (Lindl.)Lem by HPLC

摘要 目的:建立以高效液相色谱法测定络石藤中黄酮苷元成分的方法。方法:色谱柱为Dikma Inertsil ODS-3C18(250mm×4、6mm,5μm),流动相为甲醇-0.1%磷酸(pH2.5,梯度洗脱),检测波长为350nm,流速为0.8mL·min^-1柱温为32℃。进样量为10pL。结果:络石藤中黄酮类化合物多以木犀草素和芹菜素的结合形式存在,水解前后木犀草素的含量分别为0.0097%和0.0332%,芹菜素的含量分别为0.0124%和0.0709%。木犀草素的加样回收率为98.4%~102.0%,RSD为2.84%~3.42%:芹菜素的加样回收率为97.2%~101.6%,RSD为1.82%~3.98%。结论:本试验对络石藤中的黄酮苷元成分进行了初步的定性、定量研究。可为络石藤的质量研究提供科学依据。 OBJECTIVE: To determine the content of Flavonoid aglycones in Trachelosperrnurn Jasrninoides (Lindl.)Lem by HPLC. METHODS: The chromatographic separation was performed on Dikma Inertsil ODS - 3 C1s(250 mm × 4.6 mm, 5 μm) with mobile phase consisted of methnol- 0.1% phosphoric acid (pH = 2.5, gradient elution) at a flow rate of 0.8 mL · min^-1. The detection wavelength was set at 350 nm; the column temperature was 32 ℃ and the sample size was 10μL. RESULTS: The Flavanoids in Trachelosperrnurn Jasrninoides (Lindl.) Lem were mostly in combined form of Luteolin with Api- genin. The content of Luteolin before and after hydrolization was 0.009 7% and 0.033 2%, respectively, and the content of Apigenin before and after hydrolization was 0.012 4% and 0.070 9% respectively. The recovery of Luteolin was 98.4%~102.0%(RSD =2.84%~3.42%), and the recovery of apigenin was 97.2%~101.6%(RSD = 1.82%~3.98%). CONCLUSION: The quantification and qualitation of the Flavonoid aglycones in Trachelosperrnurn Jasrninoides(Lindl. )Lem was achieved elementarily in this test, and it provides scientific bases for the quality study of Trachelosperrnurn Jasminoides(Lindl.) gem.
