摘要
目的比较研究槐角不同部位,即槐角皮和种子中3种主要苷元形式黄酮类化合物,槲皮素、染料木素和山萘酚的含量差异,为科学、有效地利用现有槐角资源提供理论依据。方法采用高效液相色谱法同时测定酸水解样品中槲皮素、染料木素和山萘酚的含量。HPLC测定条件:Diamonsil C18柱(150 mm×4.6 mm,5μm),流动相为0.4%磷酸-甲醇溶液(50∶50),流速1.0 ml/min,检测波长360 nm。结果槐角皮中槲皮素、染料木素和山萘酚含量分别为(2.4±0.8)、(41.3±3.8)和(29.2±0.23)mg/g,种子中三者的含量分别为(1.9±0.4)、(3.3±1.3)和(3.6±1.1)mg/g,其中槐角皮中染料木素和山萘酚含量明显高于种子中的含量,差异显著(P<0.01)。结论槐角中主要苷元形式的黄酮类化合物为染料木素和山萘酚,且二者主要存在于槐角皮中。了解这种分布差异,可以改变以完整槐角为材料的研究思路,将槐角皮与槐角种子分别用于不同目的的研究,对于缓解目前槐角资源紧张的现状,更有效地利用现有槐资源,具有一定的指导意义。
Objective To compare the differences in quercetin,genistein,and kaempferol levels between the pericarps and the seeds of Sophora japonica L.in order to use the resources of Fructus Sophorae effectively. Methods Quercetin,kaempferol,and genistein were determined simultaneously by a method of RP-HPLC.The acidic-hydrolyzed extracts prepared from the pericarps and the seeds of Sophora japonica L.were separated on a Diamonsil C18 column(150 mm×4.6 mm,5 μm).0.07% phosphoric acid-Methanol(50:50) was used as mobile phase,the flow rate of mobile phase was 1.0 ml·min-1and the detection wavelength was set at 360 nm. Results The levels of quercetin,genistein,and kaempferol in the pericarps were(2.4±0.8),(41.3±3.8),and(29.2±0.2) mg/g,respectively,those in the seeds were(1.9±0.4),(3.3±1.3),and(3.6±1.1) mg/g,respectively.The levels of genistein,and kaempferol in the former were much higher than those in the latter(P0.01). Conclusion The main forms of flavanoid aglycone equivalents in Fructus Sophorae are genistein and kaempferol,which mainly exist in the pericarps.Being known the significant differences in the levels of genistein and kaempferol between the pericarp and the seed of Sophora japonica L.,Fructus Sophorae can be used separately in order to be used effectively.
