摘要
采用超薄等电聚焦电泳技术研究了经不同贮藏时期的两系杂交水稻F1种子中父本特征蛋白质带的变化情况。结果表明,贮藏了0~12个月的F1种子中父本特征蛋白质带依然清晰,而贮藏了18个月后,部分F1种子中的父本特征蛋白质带变得模糊。所以,贮藏一年以内的F1种子还能用超薄等电聚焦电泳技术鉴定纯度,而贮藏18个月以上的F1种子则不能继续用超薄等电聚焦电泳技术鉴定纯度。
To test the genetic purity of two-line hybrid rice F1 seeds after storage, ultrathin-layer isoelectric focusing of seed proteins (UTLIEF) was used to study the changes of the male marker protein bands (MMBs) in seeds during storage. It was found the MMBs still maintained in F1 seeds stored for 0-12 months. Howev- er, after 18 months of storage, the MMBs began to degrade and cannot be detected distinctly by using UT-LIEF. It was concluded that UTLIEF could be used to test the genetic purity of F1 seeds stored less than one year, but not of F1 seeds stored for 18 months or longer.
