摘要
目的:测定经超抗原金黄色葡萄球菌肠毒素B(SEB)和结肠癌肿瘤抗原联合修饰树突状细胞(DC)的活性。方法:结肠癌手术标本体外培养出结肠癌细胞并鉴定之,冻融法制备肿瘤抗原;同一患者外周血分离出单个核细胞,以细胞因子诱导为DC;尼龙棉柱过滤获取T淋巴细胞;肿瘤抗原和不同质量浓度的SEB联合修饰DC;以联合抗原修饰后的DC为刺激细胞,自体T淋巴细胞为效应细胞,相同时间下以不同比例共同孵育后,采用MTT法检测对结肠癌细胞的体外杀伤效果。结果:诱导出表达CD11c,CD80,HLA-DR分子的DC,经结肠癌肿瘤抗原和SEB联合修饰后,上述分子表达上调。联合抗原修饰DC的T细胞活化作用强于单独使用结肠癌肿瘤抗原修饰DC的T细胞活化作用。100μg/LSEB和肿瘤抗原联合修饰的DC以1:100比例与T淋巴细胞共孵后的杀伤肿瘤细胞效应最强。结论:SEB和肿瘤抗原联合修饰DC的活性明显强于单用肿瘤抗原修饰DC的活性。
Aim: To study the activity of dendritic cells (DC) induced by staphylococcal enterotoxin B (SEB) and tumor antigen. Methods:The cancer cells from the specimen of a patient with colon carcinoma were cultured and identified. The tumor antigen was prepared by using methods of freezing thawing. DC was obtained from peripheral blood monouclear cells and identified. T lymphocytes were acquired by nylon-fiber column filtering. Then, DC loaded with SEB and tumor antigens was hatched together with T lymphocytes by means of different proportion. The activity of T lymphocyte excited by DC was assayed with MTT method. Results: Induced DC expressed CD11c,CD80 and HLA-DR. Through modification of SEB and tumor antigen, its expression upregulated. Activation of T lymphocyte excited by DC that modified by SEB and tumor antigen was stronger than that excited by DC that modified by only tumor antigen. DC that modified by SEB and tumor antigen hatched together with T lymphocyte with a proportion had the strongest anti-tumor effects. Conclusion : The activity of DC modified by SEB and tumor antigen is stonger than the activity of DC modified only by tumor antigen.
