摘要 [目的]研究南瓜黄色素的超声波辅助提取法,并与浸取法进行比较,为开发利用南瓜提供实验依据。[方法]用超声波辅助提取法制得南瓜黄色素待测液,用Uv-2550可见紫外分光光度计在300~600nm的波长范围内测定其最大吸收波长。以提取液在特征波长处的吸光度值为指标,通过单因素实验和正交试验,确定最佳提取条件。[结果]超声波辅助提取南瓜黄色素的最佳条件为:以95%乙醇作提取溶剂,料液比(g:ml)1:10,超声波功率80W,提取时间40min,色素粗品收率为10.6%。南瓜黄色素的最大吸收波长为446nm。在超声波辅助提取南瓜黄色素的3个因素中,提取时间的R值最大,其次是超声波功率,最后为料液比。[结论]与浸提法相比,超声波提取具有高效、省时、节能的优点。 [Objective] The aim was to study the ultrasound-assisted method of pumpkin yellow pigment and compare it with leaching method so as to provide experimental foundation for exploiting and utilizing pumpkin.[Method] The ready solution of pumpkin yellow pigment was prepared by ultrasound-assisted method and its maximum absorption wavelength was determined by UV/VIS spectrophotometer Uv-250 in the wavelength range of 300~600 nm.With absorbance value of extracts at characteristic wavelength as index,the optimum extraction conditions were confirmed through single factor experiment and orthogonal experiment.[Result] The optimum conditions of extracting pumpkin yellow pigment with ultrasound-assisted method were as follows:with 95% ethanol as extraction solvent,ratio of material to liquid(g:ml) of 1:10,ultrasonic power of 80 W,extraction time of 40 min.Under the conditions,the crude yield of pigment was 10.6%.The maximum absorption wavelength of pumpkin yellow pigment was 446 nm.Among 3 factors of extracting pumpk in yellow pigment with ultrasound-assisted method,the R value of extraction time was largest,the next was that of ultrasonic power,and the least was that of ration of material to liquid.[Conclusion] Compared with leaching method,ultrasonic extraction had advantages of high efficiency,saving time and energy.
