Study on Extraction and Separation of Natural Carotene

摘要 采用浸取法从胡萝卜中提取胡萝卜素,研究了不同提取剂对胡萝卜素提取效果的影响。使用薄层色谱(TLC)、柱色谱对提取液进行分离纯化,采用紫外可见分光光度计测定胡萝卜素含量。研究结果表明:用丙酮—石油醚(3:7,V/V)混合液提取效果较好,提取的胡萝卜素含量可达995μg?g;薄层色谱分离中展开剂选用石油醚和乙醚的混合液,两者比例为9∶1时分离效果最好;柱色谱的洗脱速度和效果与溶剂极性相关,洗脱剂为石油醚和乙醚的混合液时色带分离明显。 Carotene is extracted from carrot with extraction immersion. This article studies the effect of extraction agent on the carotene extraction with the application of. Thin Layer Chromatography (TLC) and Column Chromatography are used to separate and purify the extracted carotene, and spectrophotometry is used to detect the content. The results show that the extraction with actone -petroleum ether (3:7) is better, with content as high as 995 μg/g. The separation is better when the mixture of petroleum ether and aether is chosen to be the developing agents (9:1 ). The washing speed and effect of Column Chromatography is related to the polarity of agent. The color stripe separation is clear when the mixture of petroleum ether and aether is used as washing agent.
