砍瓜×广西蜜本南瓜的杂种鉴定 被引量:3

摘要 采用聚丙烯酰胺凝胶垂直平板电泳法对砍瓜(母本)、广西蜜本南瓜(父本)以及杂种F1代的POD与Est同工酶进行测定,结果表明,父母本的两种同工酶均有明显差异,F1代两种同工酶谱表现出双亲的互补性,F1代Est同工酶出现杂种酶带Rf=0.816.证明POD和Est同工酶是可以用来鉴定南瓜品种及其杂种.气孔密度和形态也反应出双亲与F1代的差异,F1代气孔密度介于双亲之间,气孔形态偏向于父本.因此,气孔形态特征也可用来鉴定双亲和杂种.F1代瓜皮、瓜肉颜色和品质更接近父本. Isoenzymes of POD and Est of Cucurbita kangua Li. (female parent), C. moschata Duch. (male parent) and their hybrid F1 s were measured by PAGE. Meanwhile, the density and morphology of the stomata were observed. Conspicuous differences in POD and Est were observed between C. moschata Duch. and C. kangua Li, and POD and Est of the F1 s demonstrated complementarity of the parents. The hybrid F1s displayed a characteristic band of Est with an Rf of 0. 816. These results indicated that POD and Est could be used to distinguish pumpkin varieties and their hybrids. Differences also existed in the density and shape of stomata between the parents and their hybrid F1 s. Density of stomata of hybrid F1 s were intermediate between C. rnoschata Duch. and C. kangua Li. , but thire size and shape were closer to the male parent. Consequently, the parents and hybrid F1 s can be identified by comparing the density and shape of stomata. The peel, color and quality of the hybrid F1 s were closer to the male parent.
