摘要
[目的]揭示粉防己碱(Tet)对大鼠肝星状细胞(HSC)增殖的调控机理。[方法]大鼠HSC体外培养,MTT法测Tet对HSC增殖的影响,免疫细胞化学法检测Tet对HSC增殖细胞核抗原(PCNA)和细胞周期素(cyclinE)表达的影响。[结果]①Tet在0.5 mg/L~1 mg/L的范围内显著抑制大鼠肝星状细胞的增殖(F=19.364,P﹤0.05)。②0.5 mg/L、1 mg/L Tet浓度组PCNA阳性表达细胞数与对照组差异有统计学意义(F=68.329,P﹤0.O1),两浓度组之间也存在统计学差异(F=68.33,P﹤0.01)。③Tet 0.5 mg/L、1 mg/L浓度组cyclinE阳性表达细胞数与对照组差异有统计学意义(F=58.227,P﹤0.O1),两浓度组之间差异有统计学意义(F=58.23,P﹤0.01)。[结论]Tet通过抑制细胞周期素cyclinE、PCNA表达,使细胞周期停滞于G0/G1期,进而发挥抑制大鼠肝星状细胞增殖的作用。
[Objective]To study the regulation mechanisms of Tetrandrine(Tet)on the proliferation of hepatic stellate cell(HSC)and the expression of PCNA and cyclinE.[Methods]HSCs of rats were cultured in vitro in RPM1640 medium.The effects of Tet with different concentrations on the proliferation of HSCs were observed by the MTT method.While the effects of Tet on the expression of PNCA and cyclinE of HSCs were observed with immunocytochemistry.[Results]① Tet had a significant depressant effect on the proliferation of HSCs in a concentration range of 0.5mg/L-1mg/L(F=19.364,P﹤0.05).② The number of positive cells of PCNA in 0.5mg/L and 1mg/L Tet groups was significantly less than that in control group(F=68.329,P﹤0.01),and the number of PCNA positive cells was also significantly different between 0.5mg/L Tet group and 1mg/L Tet group(F=68.329,P﹤0.01).③The number of positive cells of cyclinE in 0.5mg/L and 1mg/L Tet groups was significantly less than that in control group(F=58.227,P﹤0.01),and the number of positive cells of cyclinE in 0.5mg/L Tet group was also significantly different from that in 1mg/L Tet group(F=58.227,P﹤0.01).[Conclusion]Tetrandrine can inhibit the expressions of PCNA and CyclinE of hepatic stellate cell,and make cell cycle arrest in G0/G1 stage,which may be the mechanism that tetrandrine inhibits the proliferation of hepatic stellate cells.
