糖皮质激素对雌性大鼠再生肝细胞核仁组织区的影响 被引量:3

Effects of Glucocorticoid on Nucleolar Organizer Region of Regenerating Hepatocytes in Female Rat

摘要 以肝细胞核仁组织区相关噬银蛋白(AgNORs)颗粒数目为指标,研究了大鼠部分肝切除(PH)后皮质酮对再生肝细胞转录活性的影响.结果显示:①无论何种处理方式,在PH后不同时期,肝脏门管区的肝细胞Ag-NORs数目均高于中央静脉区,大多差异极显著;②PH后24 h,各处理组(假手术、去肾上腺、去肾上腺+皮质酮)肝细胞AgNORs颗粒数均下降.除了高剂量皮质酮(40 mg/kg体重)处理组在PH 24 h后肝细胞AgNORs数持续升高外,其余各组均在PH后36 h达到最高值,随后又下降;③在PH后24~48 h,给去肾上腺大鼠注射的皮质酮剂量越高,AgNORs数目越少.以上结果表明:在PH后不同时间,大鼠肝脏门管区肝细胞的转录活性始终高于中央静脉区肝细胞;皮质酮对肝细胞的转录活性具有抑制作用,但抑制时间主要表现在PH 24 h以后. The effect of corticosterone on transcriptional activity in regenerating hepatocytes of female rats after partial hepatectomy (PH) was studied using numbers of argyrophil nucleolar organiting regions (AgNORs) as index. The result showed that: (1) numbers of AgNORs observed in portal areas were higher than that in central vein areas during liver regeneration induced by PH in all groups; (2) 24 h after PH, the numbers of AgNORs decreased whatever in sham-adrenalectomized rats, adrenalectomized rats and adrenalectomized rats treated with corticosterone. The numbers of AgNOR peaked at 36h after PH except high dose of corticosterone-treated (40 mg/kg body weight) rats (still rising); (3) treatment with corticosterone at a dose of 10, 20 or 40 mg/kg body weight respectively to the adrenalectomized rats, it would be found that the more corticosterone used, the less the AgNORs appeared. It suggested that corticosterone could inhibit transcriptional activity of regenerating hepatocytes in female rats, which happened mainly at 24 h following PH.
