猪流行性腹泻病毒S蛋白中和表位区单克隆抗体的制备与鉴定 被引量:13

摘要 以纯化的猪流行性腹泻病毒(Porcine epidemic diarrhea virus,PEDV)S蛋白中和表位(SID)重组蛋白为抗原免疫BALB/c小鼠,利用淋巴细胞杂交瘤技术获得了6株稳定分泌抗SID区特异性的单克隆抗体细胞株,分别命名为2C4、3G3、5F8、3G5、6E6和3C3。6株杂交瘤细胞诱生小鼠产生的腹水抗体效价分别为1∶51200、1∶6400、1∶12800、1∶6400、1∶51200和1∶25600。免疫球蛋白类型均为IgG1型,轻链均为Κ链。Western blot试验结果显示,6株单克隆抗体均能特异性地识别天然PEDV中的S蛋白。 The purified neutralizing epitope region of PEDV S protein, S1D, was used to immunize BALB/c mice for cell fusion. Six hybridoma cell lines secreting monoclonal antibodies (McAbs) against S1D were established and designated as 2C4, 3G3, 5F8, 3G5, 6E6 and 3C3, respectively. All six McAbs were lgG1 subtype and had K chain. The McAbs had ascetic titers of 1:51 200, 1:6 400, 1:12 800, 1:6 400, 1:51 200 and 1:25 600 respectively. Western blot confirmed that all McAbs were able to reacted with native S protein of PEDV.
