摘要
本文用同一方法在相同条件下,分别以红酵母菌株A、B、C、D、F、R发酵所得的细胞壁制备了几丁质,测定了各类菌株的几丁质含量,并用红外光谱分析法对其脱乙酰几丁质(壳聚糖)进行了比较研究。结果表明,几丁质在红酵母细胞壁中的含量比较低,不同种属红酵母菌株细胞壁中的几丁质含量差异较大。红酵母菌株D细胞壁中的几丁质含量为2.34±0.17%(w/w,干细胞),属最低,红酵母菌株C细胞壁中的几丁质含量为5.28±0.22%(w/w,干细胞),属最高。其六种壳聚糖的红外光谱基本相似,脱乙酰度相近(76%~80%),只存在个别峰值、峰位的差异,表明六种壳聚糖的基本结构相同,但其空间构象和结晶度等方面存在差异。
In this paper, different chitins were obtained respectively from Rhodotorula sp.A., Rhodotorula sp.B., Rhodotorula sp.C., Rhodotorula sp.D., Rhodotorula sp.F., and Rhodotorula sp.R., by using the same method under the same condition. The chitin content in each Rhodotorula species was determined. A comparative study was made on the deacetylation of chitin (chitosans) from several Rhodotorula species by using the infrared spectrometry. The result showed that the chitin content in the Rhodotorula cell wall was very low and the chitin content in different Rhodotorula species was different obviously. Rhodotorula sp.D. had its chitin content of 2.34 ±0.17% (w/w, dry cell) which was the least. Rhodotorula sp.C. had its chitin content of 5.28 ± 0.22% (w/w, dry cell) which was the highest. These six different chitosans had similar infrared spectra and close deacetylation degree (76% -80%). The only difference among them was in a few peaks. It was shown that their basic structures were similar and their conformation and crystallinity were slightly different.
