摘要
目的选择适宜的对照品,建立能够准确测定甘草中总黄酮含量的方法。方法针对甘草苷、柚皮苷、芦丁3种对照品分别使用紫外分光光度法,通过全波长扫描,比较不同对照品和样品与显色前后的最大吸收波长,从而确定适合的对照品。结果甘草苷对照品和样品液经过碱处理后最大吸收波长分别为334,334.5nm且全波长扫描后得到峰形基本一致,而柚皮苷对照品经过相同处理后最大吸收波长在419.5nm,芦丁对照品和样品分别经过Al2(NO3)3-NaOH-NaNO2显色后最大吸收波长在510nm和363nm。通过对3种方法的测定结果进行统计学分析,以甘草苷为对照品的测定结果分别与另外两种方法差异显著。结论选用甘草苷为对照品应用于紫外分光光度法测定甘草总黄酮成分准确度较高,是切实可行的含量测定方法。
Objective To select the appropriate standard for determination of flavonoids in Glycyrrhiza. Methods To compare the largest absorption wavelength by wavelength scanning of ultraviolet spectrophotometry. Results Standard Liquiritin and samples processed by alkali had the largest absorption at the 334nm and 334.5nm wavelength, and standard Naringin at the 419.5 nm wavelength. Conclusion To determine content of flavonoids in glycyrrhiza with ultraviolet spectrophotometry by standard Liquiritin is a practical method with higher accuracy.
